The objective of this study was to evaluate the reproductive performance, liver morphological study and post mortem characteristics of the pregnant Wistar rats treated with indomethacin, a general COX inhibitor. Indomethacin at doses of 0 (control), 0.32, 1.68 and 8.40 mg/kg/day were orally given once daily to each group (n=10) on days 3 and 4 of pregnancy (day 0 = first day of pregnancy = positive vaginal sperm). The animals were euthanized under anesthesia on day 11 of pregnancy, and were carried out necropsy and microorganism culture study. The results showed that the doses of 0.32 and 1.68 mg/kg body weight (the therapeutic dose for humans) of indomethacin caused no embryotoxic or lethal effects. In this paper we evaluated the morphologic and quantitative aspects of the implantations sites in uteruses of female pinealectomized and/or submitted to the constant illumination rats. In mammals, embryo implantation is an essential step in reproduction. Implantation is a phenomenon that involves crosstalk between the blastocyst and the maternal endometrium. However, the molecular basis of the connections between the blastocyst and endometrium is not yet fully understood. Amphiregulin is a member of the epidermal growth factor family and is known to be expressed in the luminal epithelium of the mouse uterus on 3.5 days post coitum (dpc). Rat uterine sensitization-associated gene-1 (USAG-1) mRNA is expressed in the uterus during the peri-implantation period, and its mRNA expression in uterine epithelial cells is highest on day 5 of pregnancy. On the other hand, since changes in USAG-1 mRNA expression in the mouse uterus are not seen during the estrous cycle, USAG-1 expression might be specifically regulated by embryonic factors rather than by the maternal environment. The mRNA expression patterns of EGF, HB-EGF, Amphiregulin, EGF receptor, IGF-1, CSF-1, IL-1 alpha, IL-1 beta, IL-1 receptor type 1, IL-1 receptor antagonist, LIF, COX-1, COX-2, Mucin-1, calcitonin, and rat USAG-1 mouse homologue, all of which are involved in the process of conceptus implantation to the endometrium, were examined during the estrous cycle by means of real-time quantitative PCR.
